Cette étude constitue une des trois actions engagées dans le cadre du Contrat de Plan IFREMER - Poitou-Charentes (1994-1998) sous l"intitulé Valorisation des Claires Ostréicoles. Les deux actions complémentaires concernent la compréhension et l'amélioration du verdissement des claires sous la direction du Pr. J.M. Robert de l'Université de Nantes/lSOMER et la production de masse de microalgues afin de stimuler la croissance de mollusques filtreurs sous la direction de Dr. J. Hussenot du CREMA L'Houmeau. Source/description : Genomic DNA was extracted from a whole Pacific oyster (C. gigas) after grinding in liquid nitrogen. Purified DNA was then digested by a mix of 3 restriction enzymes (AluI, RsaI, HaeIII). DNA fragments ranging from 250 to 500 bp were size-selected by agarose gel electrophoresis and ligated into the dephosphorylated blunt-ended SmaI-linearised pBKS2 plasmid (Stratagene Cloning System, CA). White colonies (11,500) were screened by hybridisation with double-stranded DNA probes containing dinucleotide repeats. Extracellular products (ECPs) of the pathogenic Vibrio aestuarianus 01/32 were previously reported to display lethality in Crassostrea gigas oysters and to cause morphological changes and immunosuppression in oyster hemocytes. To identify the source of this toxicity, biochemical and genetic approaches were developed. ECP protease activity and lethality were shown to be significantly reduced following incubation with metal chelators, suggesting the involvement of a zinc metalloprotease. An open reading frame of 1836 bp encoding a 611-aa metalloprotease (designated yam) was identified. Polyphasic analysis of four new Vibrio isolates originating from the haemolymph of diseased cultured oysters is described. The new isolates were closely related to Vibrio splendidus, having 98 % 16S rRNA gene sequence similarity. Phylogenetic analysis based on DNA gyrase subunit B (gyrB), RNA polymerase Quo factor (rpoD), replication origin-binding protein (rctB) and transmembrane regulatory protein (toxR) genes, fluorescent amplified fragment length polymorphism and DNA-DNA hybridization experiments clearly showed that the new isolates form a tight genomic group that is different from the currently known Vibrio species. Shellfish consumption is clearly associated with the transmission of enteric viruses. The channel of transmission of enteric viruses to human beings could be decreased particularly if the systems of depuration of shellfish were optimized, standardized and if their viral efficiency was controlled. The purpose of this work was to study the contamination of oysters and to assess the efficiency of several depuration processes. Our study have demonstrated that in some cases, 50% of oysters samples contain enteric viruses or H.A.V. antigen and the quantities of viruses range from 17 to 246 FF for the rotaviruses and from 124 to 200 for the enteroviruses per 100 g of tissue. Viral gametocytic hypertrophy was reported for the first time in 2001 in Pacific oyster Crassostrea gigas in France. Since this date, the number of reported cases and the distribution area have increased every year; however, the cases are not associated with macroscopic signs or increased mortality rates. This study examined the delta(15)N of suspended and sedimented particulate organic matter, primary sources of organic matter and benthic invertebrates along an estuarine gradient in Marennes-Oleron Bay, France. Particular emphasis was given to the use of delta(15)N as a tracer of the origin of organic matter and as a means of determining the benthic food web structure in estuarine environments. delta(15)N values indicated that there was direct utilisation of benthic diatoms as a food source by oyster Crassostrea gigas near intertidal mudflats but suggested trophic mediation between terrestrial detritus and oysters in the upper estuarine reaches. delta(15)N data showed that the trophic position of oysters may vary in the estuarine bay.
