Valeur énergétique de la chair de l'huître Crassostrea gigas estimée par mesures microcalorimétriques et par dosages biochimiques ArchiMer

The energetic values measured by the direct method and estimated by biochemical analysis of the annual cycle of ash-free dry weight of two Crassostrea gigas populations, show that the analytic results are inferior by 30% to those obtained by direct evaluation. The mean energetic amount is 4.84 cal.mg-1. Its seasonal variations, from 4.11 to 5.39 cal.mg-l, mainly depend on variations in the values of lipids. The mean conversion factor of lipids is 8.63 cal.mg-!, noticeably different from Brody's (9.45 caI.mg-I). This coefficient varies according to seasons from 7.85 to 9.38 cal.mg'-I. Valeurs caloriques de la chair de l'huître Crassostrea gigas Thumberg : estimation directe et biochimique ArchiMer

Caloric values of the ash-free dried sofkt parts of Crassostrea gigas have been characterized, according to season using a microcalorimeter and biochemical composition. This study shows the difference between the two methods. A conversation factor (8,63 kcal-gr(-1) has been determined for lipid of pacific oyster. Thix work shows that reproduction implies a loss of energy (53% to 63%) for non juvenile oysters. The increase of energetic level necessary to growth is more important for juvenile oysters and is upper than what is lost for reproduction.

Variabilité et bioénergétique de la reproduction chez l'huître creuse Crassostrea gigas ArchiMer

The variability in reproductive cycle of Pacific oyster, Crassostrea gigas, was evaluated in situ (Bay of Veys, BOV and Marennes-Oléron bay, MOB) and in hatchery culture conditions. Two trophic levels were tested over one year: high food level (HFL) and low food level (LFL). We observed that the phytoplanktonic concentration (chlorophyll a) was the principal factor that influenced gametogenesis intensity and duration. The oysters reared within a high trophic ration (BOV and HFL) shown a hyper-gonad development. However, the spawning efficiency was reduced and partial. Source/description : Genomic DNA was extracted from a whole Pacific oyster (C. gigas) after grinding in liquid nitrogen. Purified DNA was then digested by a mix of 3 restriction enzymes (AluI, RsaI, HaeIII). DNA fragments ranging from 250 to 500 bp were size-selected by agarose gel electrophoresis and ligated into the dephosphorylated blunt-ended SmaI-linearised pBKS2 plasmid (Stratagene Cloning System, CA). White colonies (11,500) were screened by hybridisation with double-stranded DNA probes containing dinucleotide repeats. The continuous obtention of current (speed and direction) turbidity and phytoplanktonic fluorescence was achieved for 9 successive days during the month of May 1988. Each two hours, during the day, the faeces and pseudofaeces productions of 7 oysters have been sampled individualy. The filtration rate were calculated in continuous. It appeared that the concentration in particular matter and also the planktonic fluorescence fluctuated in direct relation with the water depth, the origin of the water masses and the speed of the wind. Variations saisonnières in situ de la production et de la composition des biodépots de trois mollusques estuariens (Mytilus edulis, Crassostrea gigas, Crepidula fornicata) ArchiMer

The biodeposition was quantified on the field with sediment traps. All over the year, the biodeposition was the lowest for Crepidula fornicata. Mytilus edulis rejected more biodeposits during winter, and Crassostrea gigas during spring and autumn. The percentage of organic matter sonsumed and digested was higher during summer. The constituttive elements of the digested organic matter was not always the same, all over the year. The energy content of the digest fraction was higher in spring. For Mytilus edulis the ration intake was higher in winter and spring, related with the gametogenetic ans spawning period. An analysis of the mineral constutents of the natural food was made.

Vibrio aestuarianus zinc metalloprotease causes lethality in the Pacific oyster Crassostrea gigas and impairs the host cellular immune defenses ArchiMer

Extracellular products (ECPs) of the pathogenic Vibrio aestuarianus 01/32 were previously reported to display lethality in Crassostrea gigas oysters and to cause morphological changes and immunosuppression in oyster hemocytes. To identify the source of this toxicity, biochemical and genetic approaches were developed. ECP protease activity and lethality were shown to be significantly reduced following incubation with metal chelators, suggesting the involvement of a zinc metalloprotease. An open reading frame of 1836 bp encoding a 611-aa metalloprotease (designated yam) was identified. Vibrio gigantis sp nov., isolated from the haemolymph of cultured oysters (Crassostrea gigas) ArchiMer

Polyphasic analysis of four new Vibrio isolates originating from the haemolymph of diseased cultured oysters is described. The new isolates were closely related to Vibrio splendidus, having 98 % 16S rRNA gene sequence similarity. Shellfish consumption is clearly associated with the transmission of enteric viruses. The channel of transmission of enteric viruses to human beings could be decreased particularly if the systems of depuration of shellfish were optimized, standardized and if their viral efficiency was controlled. The purpose of this work was to study the contamination of oysters and to assess the efficiency of several depuration processes. Our study have demonstrated that in some cases, 50% of oysters samples contain enteric viruses or H.A.V. antigen and the quantities of viruses range from 17 to 246 FF for the rotaviruses and from 124 to 200 for the enteroviruses per 100 g of tissue. Viral gametocytic hypertrophy of Crassostrea gigas in France: from occasional records to disease emergence? ArchiMer

This study examined the delta(15)N of suspended and sedimented particulate organic matter, primary sources of organic matter and benthic invertebrates along an estuarine gradient in Marennes-Oleron Bay, France.
